PandaPure®️ Construct Service synthesizes genes of your protein of interest, and provides a ready-to-use DNA kit, including:

To order new PandaPure Protein Constructs, you just need to submit your proteins of interest, by providing amino acid sequence, or coding sequence, or Uniprot ID.
We recommend having a trial experiment before purifying your proteins in a large scale. For trial experiments, the smallest package (Protein Reagent PR010 and Lysis Reagent LR050) would be enough.
If you have set up a successful expression, you can scale up purification accordingly:
- 8 ml PandaPure®️ Lysis Reagent per 1 gram cells
- 30 mg proteins per 1 ml PandaPure®️ Protein Reagent
Here is a rule of thumb to choose package sizes.
Per 1 liter bacterial culture, you will have ~ 12 gram cells, and need:
- 100 ml PandaPure®️ Lysis Reagent
- 20 ml PandaPure Protein Reagent

While PandaPure significantly enhances the capability to express a wide range of proteins, including many that are traditionally challenging, it does not guarantee high-yield, high-purity protein production on the first attempt. The success of recombinant protein production, particularly in terms of yield and purity, largely depends on the inherent properties of the target protein, such as solubility, conformation, and thermostability. Additionally, variations in bacterial culture conditions across different laboratories may affect reproducibility.
For optimal outcomes, we recommend conducting preliminary small-scale tests using the PandaPure workflow to evaluate expected yield and purity before proceeding to larger scale applications. This approach helps in assessing the efficiency of PandaPure for your specific protein targets.
To further support your protein production success, every gene is also cloned into pET28a(+), a widely utilized traditional vector, as a control. If a protein fails to express in both the PandaPure system and with pET28a(+), it suggests that the protein may be exceptionally challenging to produce, indicating a potential need for further protein engineering. For such cases, we offer custom projects through Ailurus Protein Service, which includes AI-aided design and high-throughput screening to enhance protein expression outcomes.
Should your gene not express in the PandaPure system but successfully does so with pET28a(+), you have the flexibility to revert to the traditional expression system. This ensures you have a backup plan and further emphasizes our commitment to supporting your research needs with adaptable solutions.
You can replace PandaPure Lysis Reagents by other non-ionic detergents, such as B-PER from Thermo Fisher Scientific, and Bugbuster from Merck Millipore.
However, you cannot use alkaline lysis reagents from plasmid mini-prep kits to perform PandaPure.
They cannot, unfortunately. PandaPure technology requires the host cells to do a lot of works intracellularly, which are encoded in specific DNA sequences from PandaPure Protein Construct.
You can just submit the sequence, and we'll proceed the order as usual.
In fact, nowadays, it is cheaper to resynthesize the gene than the shipping the genes, unless you have a large protein bigger than 1000 amino acids.
Of course, you can also choose to send us your plasmids for subcloning. Extra fees for shipping, handling, and sequencing of the materials will apply, according to the real expenses.
You only need basic equipment for molecular biology, such as incubator and shaker to grow bacteria culture, water bath or thermocycler, and centrifuges. You also need materials for bacterial transformation, including competent host cells (either chemical or electro-compent celle works), medium for growth, and basic glasswares. To induce the expression, you would also need two chemical inducers, Isopropyl β-D-1-thiogalactopyranoside (IPTG) and anhydrotetracycline (aTc). For high purity production, you also need lysozyme to better disrupt bacterial cell walls.
PandaPure technology supports most of the strains of E. coli, if a competent cell can be made.
By default, our design of constructs uses T7 promoters to drive gene expression, thereby requiring T7 polymerase-encoding hosts, such as BL21(DE3) and BL21AI. Also, these constructs contain kanamycin (Kan) and chloramphenicol (Cm) resistances, and cannot propagate in hosts with the same markers.
If you want to perform PandaPure in non-T7 hosts, or hosts that are already KanR+ or CmR+, please let us know the details of your host strain, to modify the constructs accordingly in advance.
