PandaPure® Protein Reagent induces efficient and precise tag cleavage within organelles, and mediates the release of target proteins. By default, these PandaPure Protein Kits provide HEPES buffered PandaPure Protein Reagent (pH ~ 8), with 100 mM NaCl for protein stability. Additional components such as protease inhibitors, salts, and reducing agents may be added, depending on the applications.
The recommended amount of PandaPure® Protein Reagent is 2-10 ml per gram of cells. As a rule of thumb, a 50 ml overnight culture usually produces about 0.5-0.7 g cell pellets, and requires ~ 5 ml reagents. For small cultures, 1/5 culture volume of reagents can be used for resuspension. For high-throughput practices, larger volume can be used for simple liquid handling. To be noticed, adding too much Protein Reagent will dilute the sample.
The optimal volume of Protein Reagent used for your experiment is based on the amount of protein expressed. Thus, we suggest starting a small-volume experiment first, before you scale up the production.
PandaPure Protein Reagent Hi, is a variant of PandaPure Protein Reagent, optimized for high-throughput experiment, enabling higher efficiency and higher purity. Notably, this variant contains more chemicals that may affect downstream applications - therefore we recommend diluting the purified product before using it.

To construct the expression plasmid, 25-250 ng linearized expression vector is often needed per cloning, depending on the method.
For a quick launch, we recommend > 10 ng expression plasmid if transformed into a competent cell already containing the related organelle, or >250 ng organelle and expression plasmid each for co-transformation.
Ailurus PandaPure® Lysis Reagent (in 1 M Tris-HCl, pH 7.5) is an easy-to-use bacterial lysis reagent based on nonionic detergent. PandaPure® Lysis Reagent enables gentle lysis of bacteria (E. coli) without mechanical disruption. It can also be used for washing impurities in PandaPure workflows. Lysozyme and DNase I can be further supplied to improve extraction efficiency of large proteins (MW > 70kDa).
The reagents, like many alternative commercial products, are compatiable with other downstream applications, such as affinity chromatography, SDS-PAGE, and protein quantification. Protease inhibitors, RNase inhibitors, salts, reducing agents, denaturants and other components may be added to the reagent for particular applications.
The recommended amount of PandaPure® Lysis Reagent to use is 8 ml per gram of cells. A 50 ml overnight culture, which usually produce about 0.5-0.7 g cell pellets, needs 5 ml reagents. For small cultures, 1/5 culture volume of reagents can be used for resuspension. For high-throughput practices, larger volume can be used for simple liquid handling. To be noticed, adding too much lysis reagent may extract target proteins into the supernatant prematurely, and result in lower yield of final product.
