Plasmid systems

Organelle plasmids encode the formation of the functional synthetic organelles by phase separation and self-assembly of biomolecules, e.g. synthetic RNA organelle designed under the architecture of Transcriptionally Engineered Addressable RNA Solvents (TEARS).
  • pOrganelle(pTEAR-2): synthetic RNA organelle, TEARS architecture, MS2 RNA aptamer as its receptor, PLtetO-1 promoter (aTc inducible), p15A origin of replication, chloramphenicol resistance (CmR)
The compatible expression plasmid need to encode protein of interest (POI) fused with cognate signal tag to address the chosen synthetic organelle.
  • pPPC-vector: designed for Gibson assembly cloning, MS2 coat protein derived signal tag for C-terminal fusion, pET/pBR322 origin of replication, kanamycin resistance (KanR).
To construct the expression plasmid, 25-250 ng linearized expression vector is often needed per cloning, depending on the method.
For a quick launch, we recommend > 10 ng expression plasmid if transformed into a competent cell already containing the related organelle, or >250 ng organelle and expression plasmid each for co-transformation.
